چکیده

The b 2 integrins and intercellular adhesion molecule-1 (ICAM-1) are important for monocyte migration through inflammatory endothelium. Here we demonstrate that the integrin a v b 3 is also a key player in this process. In an in vitro transendothelial migration assay, monocytes lacking b 3 integrins revealed weak migratory ability, whereas monocytes expressing b 3 integrins engaged in stronger migration. This migration could be partially blocked by antibodies against the integrin chains a L, b 2, a v, or IAP, a protein functionally associated with a v b 3 integrin. Transfection of b 3 integrin chain cDNA into monocytes lacking b 3 integrins resulted in expression of the a v b 3 integrin and conferred on these cells an enhanced ability to transmigrate through cell monolayers expressing ICAM-1. These monocytes also engaged in a L b 2-dependent locomotion on recombinant ICAM-1 which was enhanced by a v b 3 integrin occupancy. Antibodies against IAP were able to revert this a v b 3 integrin-dependent cell locomotion to control levels. Finally, adhesion assays revealed that occupancy of a v b 3 integrin could decrease monocyte binding to ICAM-1. In conclusion, we show that a v b 3 integrin modulates a L b 2 integrin-dependent monocyte adhesion to and migration on ICAM-1. This could represent a novel mechanism to promote monocyte motility on vascular ICAM-1 and initiate subsequent transendothelial migration.

N-acetylglucosaminyltransferase V negatively regulates integrin α5β1-mediated monocyte adhesion and transmigration through vascular endothelium.

Changes in the expression of glycosyltransferases that branch N-linked glycans are associated with many physiological and pathological events, such as cell adhesion, migration, proliferation and tumor cell malignancy. Here, the altered levels of N-acetylglucosaminyltransferase V (GnT-V) and its product β(1,6)-linked GlcNAc in monocytes were observed during inflammation. The effects of GnT-V and...

Molecular mechanisms of monocyte adhesion to interleukin-1beta-stimulated endothelial cells under physiologic flow conditions.

This study identifies multiple pathways used by monocytes to adhere to 4-hour interleukin-1beta stimulated human umbilical vein endothelial cells under flow conditions. Physiologic shear stresses were simulated in a flow chamber with parallel plate geometry; quantitation of primary adhesion, secondary adhesion, and transmigration was performed using phase contrast videomicroscopy. Neuraminidase...

RhoA is required for monocyte tail retraction during transendothelial migration

Transendothelial migration of monocytes is the process by which monocytes leave the circulatory system and extravasate through the endothelial lining of the blood vessel wall and enter the underlying tissue. Transmigration requires coordination of alterations in cell shape and adhesive properties that are mediated by cytoskeletal dynamics. We have analyzed the function of RhoA in the cytoskelet...

Monocyte-selective transendothelial migration: dissection of the binding and transmigration phases by an in vitro assay

We describe a quantitative assay of transendothelial migration (TEM) that allows us to selectively study the interaction of monocytes with confluent human endothelial cell (HEC) monolayers. The HEC are grown on hydrated collagen gels; the monocytes need not be purified. 100% of monocytes transmigrated the monolayer within 1 h at 37 degrees C and accumulated in the subendothelial collagen; TEM o...
